Abstract

In this study, we evaluated the polyphenol content, flavonoid content, antioxidant and biological of 70% EtOH extract and its fractions (dichloromethane, ethyl acetate and water) of the plant Chrysanthemum indicum L. (CI) belonging to family chrysanthemum. antioxidant activity was determined 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2’-azino-bis-3-ethylbenzthiazoline-6-sulphonic acid (ABTS), and superoxide scavenging activity. The biological activity tests included measurement of α-glucosidase and PTP1B. The results show that content of polyphenol and flavonoid in 70% EtOH of CI 30.20 mg and 31.58 mg, respectively. Among the different samples, ethyl acetate (EA) fraction showed the highest level of polyphenol (87.87 mg) and flavonoid (65.12 mg). The DPPH, ABTS and superoxide dismutase(SOD) antioxidant assay of EA fraction showed 93.84%, 93.38% and 84.68% inhibition respectively. The EA fraction also showed the highest antioxidant activity compared to 70% EtOH extract and other fractions. EA fraction (20 μg/mL) showed the highest 30.29% and 99.24%, respectively compared to other samples. The results of this study show that the EA fraction contained more antioxidant and antidiabetic components than 70% EtOH extract and other fractions. In conclusion, we believe that CI may be useful natural antioxidant and functional material.

서 론

감국(Chrysanthemum indicum L.)은 관속식물의 국화과 국화속에 속하는 국내 자생의 여러해살이 초본으로, 높이는 30∼60 cm이고, 잎은 긴 타원형-난형이며, 깃털 모양으로 다섯 갈래로 갈라진다. 9∼10월에 노란 꽃이 피며 이 꽃을 건조한 것이 감국이라고 불리고 있다(Yoon OH & Cho JS 2007). 황국(黃菊)이라고도 불리는 감국에는 휘발성 정유성분이 함유되어 있으며, 각종 비타민들이 들어있고, 한방에서는 꽃부분에 감미(甘味)가 있어 감국(甘菊)이라 하며, 약용적인 부분 이외에도 좋은 향기로 인하여 국화주, 향로 등으로 이용하기도 하였다(Park JH & Lee JK 2000). 중국에서는 감국을 장수식품으로 여겨 감국주를 담아 먹거나, 감국차로 마시기도 한다(Shin YJ 등 2004). 이러한 감국을 한의학에서는 소염, 해열, 두통 완화, 혈압 강하 등으로 이용되고 있으며(Park CS 1965), 최근 여러 연구들을 통해 감국의 항균(Jang DS 등 1999), 항염(Shunyaing Z 2005; Lee DY 2009), 면역조절 효과(Heon MS 2009; Flen AN 1999; Kong LD 2000; Wang ZG 2000)와 항암활성(Cheng W 2005) 등이 밝혀지고 있다. 또한, 감국은 최근 미용의 용도로 여드름과 보습효과가 있다고도 알려져 있다. 이러한 감국의 화학적 성분에 대한 연구로는 luteolin, apigenin, acacetin 및 flavonoid 배당체들(Ryu SY 1994; Chatterjee A 1981), lactone류(Chen Z & Peijuan X 1987), 정유(Uchio Y 등 1981), sesquiterpene이 생약학적 측면에서는 많은 연구가 수행되고 있다(Mladenova K 등 1988; Jung KY 등 1996). 그러나 추출방법에 따른 연구나 용매에 대한 연구는 많이 진행되어 있으나, 분획을 통한 분획물에 대한 연구는 미비한 상태이다. 이에, 본 연구에서는 감국 추출물과 분획물을 이용하여 항산화 효능과 생리활성에 대해서 확인해 보고자 하였다.


재료 및 방법
1. 재료 및 시료추출

본 실험에 사용된 감국(Chrysanthemum indicum L; CI)은 건강보감(충북 제천)에서 구매하여 사용하였다. 건조된 CI 50 g에 10배의 70% ethyl alcohol(70% EtOH)을 가하여 80℃에서 2시간 2회 환류 추출을 진행하였다. 추출한 후 여과지(Whatman No.1, MO, USA)를 이용하여 여과하였고, rotary evaporator(EYELA SB-1000, Tokyo, Japan)를 이용하여 농축하였으며, CI 70% EtOH 50g을 증류수에 현탁하고, 동량의 dichloromethane(DM)과 함께 분획 깔때기에 넣고 충분히 섞어 주고, DM를 수득하는 과정을 3회 반복하였다. DM을 전부 모아서 raotary evaporator에서 농축하여 DM fraction(20 g, 수율 40%)을 얻었다. 남은 수층에 ethyl acetate(EA)를 가하여 분획 깔때기에서 분획하는 과정을 3회 반복하여 EA를 얻은 다음 이를 모아 rotary evaporator에서 aspirator로 감압하여 용매를 제거하여 EA fraction(11 g, 수율 22%)을 얻었다. 최종적으로 남은 층(H2O) 또한 rotary evaporator에서 aspirator로 감압하여 물을 제거한 후, H2O fraction(19 g, 수율 38%)을 얻었다. 이후, 추출물 및 분획물을 동결건조(Operon FDU-8606, Gimpo, Korea)를 진행한 후 본 실험에 사용하였다.

2. 실험 시약

본 실험에 사용된 시약은 Folin-Ciocalteu's phenol reagent, tannic acid, microplate reader, AlCl3 ethanol solution, quercetin, 1,1-diphenyl-2-picrylhydrazyl, 2,2’-azino-bis-3-ethylbenzthiazoline-6-sulphonic acid, tris-HCl, phenazine methosulfate, nitro blue tetrazolium, β-nicotinamide adenine dinucleotide Saccharomyces cerevise로부터 얻어진 α-glucosidase, p-nitrophenyl-α-D-glucopyranoside(pNPG), p-nitrophenyl phosphate disodium salt hexahydrate(pNPP), ethylene diamine tetraacetic acid(EDTA), dithiothreitol(DTT), acarbos(ACA), ursolic acid (UA) 등은 모두 Sigma Chemical Co.,St. Louis, MO, USA에서 구입하였으며 protein tyrosine phosphatase 1B는 R&D Systems (Minneapolis, MN, USA)에서 구매하였다.

3. 총 폴리페놀 및 플라보노이드 함량

총 폴리페놀 함량은 Folin-Denis법을 변형하여 측정하였다(Folin O & Denis W 1912). CI 70% EtOH 추출물(4 µg/mL, 20 µg/mL, 100 µg/mL)과 분획물(0.8 µg/mL, 4 µg/mL, 20 µg/ mL)을 0.2 mL씩 취하고, 여기에 Folin-Ciocalteu's phenol reagent 0.2 mL를 첨가하여 잘 혼합한 후, 3분간 실온에 방치하였다. 3분 후 10% Na2CO3 0.4 mL를 가하여 혼합하고, 증류수를 4 mL 첨가한 후 차광하여 실온에 1시간 방치한 후, 725 nm에서 microplate reader(Molecular Spectramax340, CA, USA)를 측정하였다. 이때 총 폴리페놀 화합물은 tannic acid를 이용하여 작성한 표준곡선으로부터 함량을 구하였다. 총 플라보노이드 함량은 Zhuang의 방법을 변형하여 측정하였다(Zhuang XP 등 1992). 즉, CI 70% EtOH 추출물과 분획물을 농도별로 0.1 mL씩 넣고 2% AlCl3 ethanol solution 0.1 mL를 가하여 실온에서 1시간 방치한 후, 420 nm에서 흡광도를 측정하였다. 표준물질로는 quercetin을 사용하였으며, 표준물질의 검량선과 비교하여 총 플라보노이드 함량을 구하였다.

4. DPPH 및 ABTS 라디칼 소거 활성

1,1-Diphenyl-2-picrylhydrazyl(DPPH) 라디칼 소거능은 Blois의 방법을 변형하여 DPPH 라디칼에 대한 시료 용액과의 전자공여효과로 반응에 의해 DPPH radical이 감소하는 정도를 흡광도로 측정하였다(Blois ML 1958). CI 70% EtOH 추출물과 분획물을 농도별로 0.5 mL 취하고, 0.1 mM DPPH solution 1 mL를 가하여 vortex 후 암실상온에서 30분간 반응시켜 517 nm에서 흡광도를 측정하였다.

2,2’-Azino-bis-3-ethylbenzthiazoline-6-sulphonic acid(ABTS) 라디칼 소거능은 Re의 방법에 준하여 전자 공여 효과로 반응에 의해 라디칼이 감소하는 정도를 확인하는 실험을 진행하였다(Re R 등 1999). 7.4 mM의 ABTS + 2.6 mM potassium persulfate를 24시간동안 실온에서 라디칼을 형성한 후, 실험 직전 ABTS 용액을 microplate reader를 이용하여 흡광도가 0.70±0.03(mean±S.D.)이 되도록 phosphate-buffer saline(pH 7.4)을 이용하여 희석하여 맞추어 주었다. 이후 CI 70% EtOH 추출물과 분획물을 0.1 mL에 ABTS용액 0.9 mL를 가하여 실온암소에서 10분간 반응시킨 후 732 nm에서 흡광도를 측정하였으며, 계산식은 아래와 같다.

5. Superoxide Radical 소거능

Superoxide radical 소거 활성 측정은 Nishikimi의 방법에 따라 다음과 같이 측정하였다(Nishikimi M 등 1972). CI 70% EtOH 추출물과 분획물을 농도별로 0.5 mL씩 넣고 0.1 M Tris-HCl 완충용액(pH 8.5) 0.1 mL, 100 uM phenazine methosulfate(PMS) 0.2 mL를 혼합하여 반응시킨 후, 500 µM Nitro blue tetrazoliuym(NBT) 0.2 mL 및 500 µM β-nicotinamide adenin dinucleotide(NADH) 0.4 mL를 첨가하여 실온에서 10분간 반응시킨 다음 microplate reader를 이용하여 560 nm에서 흡광도를 측정하였다. Superoxide radical 소거능성은 추출물의 첨가 전과 후의 차이를 아래와 같이 백분율로 나타내었다.

6. α-Glucosidase 저해 활성

Kim의 방법에 따라 CI 분획물(0.8 µg/mL, 4 µg/mL, 20 µg/ mL)을 8 µL, 0.2 unit/mL α-glucosidase 20 µL와 0.1 M potassium phosphate buffer(pH 6.8) 112 µL를 잘 혼합하여 37℃에서 10분간 전처리하였다(Kim KY 등 2008). 이후 1.25 mM pNPG(p-nitrophenyl α-D-glucopyranoside) 20 µL를 첨가하여 37℃에서 10분간 반응시킨 후 2 M Na2CO3 80 µL로 반응을 정지시켜 405 nm에서 흡광도를 측정하였다. 이때, 활성 비교를 위하여 대조약으로 acarbose(50 μg/mL)를 사용하였다. 저해활성률은 아래 식과 같이 산출하였다.

7. Protein Tyrosine Phosphatase 1B(PTP1B) 저해활성

Tonks의 방법에 따라 pNPP를 기질로 이용하여 탈인산화 정도를 측정하여 PTP1B에 대한 저해활성을 조사하였다(Tonks N 등 1988). CI 분획물(0.8 µg/mL, 4 µg/mL, 20 µg/mL)을 20 µL에 distill water에 녹인 PTP1B(0.5 U/mL) 40 µL를 넣고 2 mM pNPP(0.1 M NaCl, 1 mM EDTA, 50 mM 1 mM DTT) 50 µL와 혼합한 후, 37℃에서 30분간 반응시킨다. 10 N NaOH를 100 µL 첨가하여 반응을 종결시킨 다음 405 nm에서 흡광도를 측정하였다. 이때, 활성 비교를 위하여 대조약으로 ursolic acid(2 μg/mL)를 사용하였다. 저해활성률은 아래 식과 같이 산출하였다.

8. 통계처리

통계 분석은 IBM SPSS Statistics(Version 21.0. Armonk, NY)을 이용하여 각 측정군의 평균과 표준편차를 산출하고, 처리간의 차이 유무를 one-way ANOVA로 분석한 뒤, Duncan’s multiple range test를 이용하여 p<0.05 수준에서 통계 분석을 진행하였다.


결과 및 고찰
1. 총 폴리페놀 및 플라보노이드 함량

항산화물질로서 예방효과가 증명된 각종 비타민들과 페놀산, 플라보노이드 등의 식물성 폴리페놀과 같은 천연 항산화제에 대한 연구가 진행되고 있다(Kim WK 등 1999; Kim NE & Kim WK 1999; Terao J 1989). CI 추출물 및 분획물의 총 폴리페놀 함량은 EA fraction이 표준물질 검량선에 의해 7.87 mg/mL로 가장 높은 함량을 보였으며, DM fraction이 33.46 mg/mL, 70% EtOH가 30.20 mg/mL, H2O fraction 28.66 mg/ mL 순으로 확인할 수 있었다(Table 1). 이는 CI에 함유된 많은 정유 성분들이 분획과정에서 EA layer에 집중한 것으로 보이나, 70% EtOH에서의 폴리페놀 함량이 많지 않은 것은 Kim JY 등(2007)의 연구를 통해 유기용매 분획물의 페놀성 화합물은 ethyl acetate fraction > butanol fraction > dichloromethane fraction > ethanol extract > aqueous fraction > hexane fraction 순으로 나타나며, 특히 ethyl acetate fraction에서 페놀성 화합물이 가장 높게 나타난다는 것으로 확인이 가능하다. 총 플라보노이드 함량 역시 EA 분획물에서 표준물질 검량선에 의해 65.12 mg/mL로 가장 높은 성향을 보였으며, DM fraction이 34.80 mg/mL, 70% EtOH가 31.58 mg/mL, H2O fraction이 30.29 mg/mL로 나타나는 것으로 보아 폴리페놀과 Kim 등(2007)의 결과와 일치하는 모습을 보였다.

2. DPPH 및 ABTS 라디칼 소거 활성

Kim(Kim JH & Kim MK 2003)에 의하면 전자공여능은 활성 라디칼에 전자를 공여하여 식품 중의 지방질 산화를 억제하거나, 인체 내에서는 활성 라디칼에 의한 노화를 억제시킬 수 있는 능력을 의미한다. 감국의 각 추출물 및 분획물의 DPPH 및 ABTS radical 소거활성을 측정하였고, 그 결과는 Table 2와 같다. DPPH radical 소거활성의 경우, EA fraction이 20 μg/mL의 농도에서 93.84%의 소거능을 보이며, DM fraction은 20 μg/mL에서 42.10%, 70% EtOH이 100 μg/mL에서 40.04%, H2O fraction이 20 μg/mL 37.21%로 나타났다. 그 중 EA fraction에서 가장 높은 소거능을 보여주고 있으며 ABTS radical 소거활성의 경우도 EA fraction의 20 μg/mL의 농도에서 93.84% DM fraction이 20 μg/mL의 농도에서 34.80 %, 70% EtOH이 100 μg/mL의 농도에서 40.04%, H2O fraction이 20 μg/mL의 농도에서 37.21%로 나타났다. DPPH와 ABTS radical 소거활동 모두 EA fraction에서 가장 효능이 좋게 나왔다. Ahn MS 등(2007)의 연구에서 총 폴리페놀 함량과 자유 라디칼 소거활성이 높은 상관성이 있다고 보고하였으며, 또한 Lee GD 등(1997)의 연구에서도 폴리페놀과 자유 라디칼 소거활성에 대한 연관성을 보고하고 있으며, 이는 Anagnnostopoulou MA 등(2006)의 연구를 통해 DPPH 라디칼 소거활성이 페놀성 물질에 의한 항산화작용의 지표라는 연구결과와 밀접한 관련이 있다고 보며, 본 연구의 결과도 총 플라보노이드와 폴리페놀, 자유 라디칼 소거활성의 결과가 동일하다.

3. Superoxide Radical 소거능

노화와 질병의 원인 중의 하나로 자유 라디칼을 억제하는 작용으로는 전자공여작용(DPPH, ABTS), Superoxide dismutase(SOD) 유사활성 등이 있다. Valko M 등(2007)에 의하면 전자공여작용은 자유 라디칼에 전자를 공여하여 산화를 억제하는 것을 말하고, SOD는 생체 내에서 생성되고 전자 환원으로 파괴성이 큰 초과산화물 음이온기를 정상상태의 산소로 전환시켜주는 것으로 알려져 있다. CI 추출물 및 분획물을 비효소적 방법인 NADH/PMS로 유발된 Superoxide radical 소거능을 측정하였고, 그 결과 CI 70% EtOH의 농도별(4 μg/mL, 20 μg/mL, 100 μg/mL)의 소거활성은 농도 의존적으로 나타났으며, 이는 대조군인 ascorbic acid(20 μg/mL)보다 모두 유의적으로 나타났다(Fig. 1). 분획물의 경우, EA fraction이 20 μg/mL의 농도에서 93.84%의 소거능을 보였으며, DM fraction이 20 μg/mL 농도에서 42.10%, H2O fraction이 20 μg/mL의 농도에서 37.21%로 나타났다(Fig. 2). EA fraction의 경우, 대조군인 ascorbic acid(20 μg/mL)보다 2배 이상 소거활성을 보이는 것으로 확인된다. 이러한 결과는 전자공여작용인 DPPH나 ABTS와 같은 결과 값을 보여주어 SOD 소거활성의 경우에도 페놀성 물질에 의한 항산화 작용이라고 볼 수 있다.

4. α-Glucosidase 저해 활성

α-Glucosidase는 체내에 저장되어 있거나 혈중에 존재하는 다당류를 단당류로 분해하는 역할을 한다(Lee BH 등 2012). 그러므로 α-glucosidase 효소를 억제함으로써 혈당치를 저하시켜 당뇨 발생을 막을 수 있다(Kim SS 등 2009). 이와 같은 당뇨억제활성을 확인하기 위해 CI 추출물을 대상으로 α-glucosidase 효소 억제율을 확인하였다. α-Glucosidase 활성 저해 효과를 측정한 결과, EA fraction을 농도별(0.8 μg/mL, 4 μg/ mL, 20 μg/mL)로 처리하였을 때 각각 9.27%, 17.71%, 30.29 %로 α-glucosidase의 활성 저해효과를 보였으며, 대조군 acarbose(50 μg/mL)는 50.12%의 억제율을 나타내었다(Fig. 3). 위 실험의 결과에서 대조군 acarbose는 α-glucosidase 저해제로 소장내의 점막에서 다당류를 분해하는 α-glucosidase를 억제함으로써 탄수화물의 소화흡수를 저해함으로써 식후 혈당을 낮춰주는 것으로 보고되었으며(Martin AE & Montgomery PA 1996), EA fration이 acarbose와 비슷한 결과를 보임으로써 α-glucosidase 활성 억제 작용이 있다고 확인할 수 있다.

5. Protein Tyrosine Phosphatase 1B(PTP1B) 저해활성

인슐린이 그 수용체와 결합하면 IRS-1(Insulin receptor substrate-PI3K(Phosphoinositide 3-kinase) 경로를 통해 당 대사와 글리코겐 합성을 유도하게 된다. 이때 인슐린과 인슐린 수용체의 결합을 방해하는 효소가 PTP1B이며, PTP1B의 활성화를 억제하면 인슐린의 작용을 유도하게 되므로 항당뇨 효과를 확인할 수 있다(Elchebly M 등 1999). PTP1B 활성 저해효과를 측정한 결과, 대조군 ursoric acid(2 μg/mL)에 비해 DM fraction을 0.8 μg/mL, 4 μg/mL, 20 μg/mL로 처리 시 각각 42.55 %, 52.66%, 92.45%, EA fraction은 각각 84.1%, 90.78%, 99.24 %로 통계적으로 유의하게 PTP1B의 활성 저해효과를 보였다(Fig. 4). 앞서 α-glucosidase 실험 결과, EA fraction에서 억제 효능을 확인하여 현재 당뇨와 비만 치료제로 알려진 PTP1B 억제 효능을 확인하였다. Cl 분획물의 PTP1B 억제 효능을 확인한 결과, α-glucosidase 억제 결과와 같이 EA fraction에서 억제 효능이 높게 나타났다. 이는 EA fraction 속에 α-glucosidase와 PTP1B를 억제하는 다량의 활성 성분들이 효능을 나타내었을 것으로 추측할 수 있으며, Cl 분획물이 α-glucosidase 억제함으로써 혈당조절에도 관여하고, PTP1B 억제를 통해 인슐린 신호전달에도 관여함으로써 항당뇨 효능을 보여주는 것으로 확인되었다.

요 약

본 연구에서는 국화과에 속하는 Chrysanthemum indicum L.(CI)의 70% EtOH 추출물 및 분획물(dichloromethane, ethyl acetate 및 H2O)의 폴리페놀 함량, 플라보노이드 함량, 항산화 활성 및 생리활성을 평가하였다. 항산화 활성으로는 DPPH, ABTS 및 superoxide 소거 활성을 확인하였고, 생리활성 평가는 α-glucosidase와 PTP1B 저해 활성을 측정하였다. 그 결과, CI 70% EtOH 추출물에서 폴리페놀과 플라보노이드의 함량은 각각 30.20 mg 및 31.58 mg인 것으로 확인되었다. 여러 분획물 중 EA 분획물이 폴리페놀(87.87 mg)과 플라보노이드(65.12 mg)의 함량이 가장 높았다. EA 분획물의 DPPH, ABTS 및 SOD 소거능은 각각 93.84%, 93.38% 및 84.68%의 억제를 나타냈다. EA 분획물은 70% EtOH 추출물 및 다른 분획과 비교하여 가장 높은 항산화 활성을 보였다. 또한 EA 분획물(20 μg/mL)의 α-glucosidase 및 PTP1B 억제 결과는 다른 시료에 비해 각각 30.29% 및 99.24%의 가장 높은 억제를 나타내었다. 본 연구 결과를 통해 감국의 EA 분획물이 70% EtOH 추출물 및 다른 분획물보다 많은 항산화 및 항당뇨 성분을 함유하는 것으로 보여진다. 결론적으로 우리는 CI의 천연 항산화 물질 및 기능성 물질에 성능이 좋아 화장품 첨가제나 음료로도 개발이 가능할 수 있을 것으로 사료된다.
